At
present we have four instruments in the Analytic and
Preparative Cytometry Core:
Introducing the
newest addition to our instrumentation:
BD LSRII Custom Designed Flow
Cytometer
This instrument is equipped with Blue
488nm, Green 532nm, Red 633nm, and Violet 405nm lasers.
The octagon and trigon optical arrays will enable
detection of 2 colors off the blue, 5 colors off the
green, 3 colors off the red, and 8 colors off the violet
laser. Dichroic mirrors and bandpass filters are
configured to detect the following dyes:
Blue laser: FITC, Alexa 488, PE
Green laser: PE CY7, PE CY5.5,
PerCP CY5.5, PE Texas Red, PE
Red laser: APC, Alexa 700, APC CY7
Violet laser: Pacific Blue, AmCyan,
Q dots
The acquisition/ analysis software is DiVa (PC based).
Data can also be saved in FCS format and analysed using
FlowJo software.
If a laboratory is interested in trying the instrument
out, we suggest that one person be designated as the
trainee. They will in turn train other members of the
lab. We have identified some people who are familiar
with the software. We will arrange training sessions as
the need arises.
We also continue to
maintain:
FACScan

This instrument is
manufactured by Becton-Dickinson and contains a 488 nm
argon laser. It can detect
up to three colors
simultaneously such as FITC, PE, and PerCP. It is
connected to a MacIntosh computer
and utilizes "CellQuest" as
the acquisition and analysis software.
This software is
user-friendly and our
facility offers tutorials
for those interested in learning to acquire/analyze
their own samples. Please call 934-1362 or email
Enid.Keyser@ccc.uab.edu
to schedule your tutorial.
FACSCalibur
This instrument is also
manufactured by Becton-Dickinson and contains two
lasers. A 488 nm argon and
a 633nm red diode laser
enables this instrument to detect up to four colors
simultaneously such as FITC,
PE, PerCP, and APC. The
FACSCalibur is also linked to a MacIntosh computer and
uses "CellQuest" software. If you would like more
information on running four color experiments please
call 934-1362 or
email
Enid.Keyser@ccc.uab.edu.
FACSVantage SE with
DiVa option

This instrument is also
manufactured by Becton-Dickinson and has cell sorting
capabilities. It is
equipped with two lasers, one coherent which emits at
488 nm and has a UV line which emits at 360 nm,
and one which is a HeNe
emitting at 633 nm. With the addition of the DiVa
option we are capabale of running eight colors plus
forward and side scatter parameters, as well as 2-way
and 4-way sorting. For more information on setting up a
sorting experiment please call 934-1362 or email
Enid.Keyser@ccc.uab.edu.
